This thesis reports time resolved polarized optical microscopy (POM), light scattering (DLS) and high and low X-ray scattering (SAXS/WAXS) experiments used for the study of G-Hydrogel formation kinetics, a particular system of biotechnological interest. G-Hydrogels are gels formed with the combination of Guanosine and GMP (Guanosine 5’ monophosphate) molecules in water, wich leads to structures called quadruplexes, these structures are able to form a stable 3D network that can absorb large volumes of water. Due to their biocompatibility these gels are useful in many applications such as Drug Delivery Systems or protein storage. In order for a possible use of this system in Local Drug Delivery, wich requires gel formation in situ at body temperature, the gel formation and propagation kinetics were been studied. The results in laboratory from POM and DLS show that gel formation is faster at low water concentrations, while slower at high water concentrations. At the same time thanks to SAXS and WAXS experiments carried out at the Diamond Light Source synchrotron in Oxford and Elettra syncrotron in Trieste it has been observed that the quadruplexes structure can be different according to Guanosine and GMP ratio. These results are important for understanding the behavior of the gel when the Guanosine and GMP solutions are injected separeted.

Questa tesi riporta l'utilizzo di tecniche di microscopia ottica a luce polarizzata (POM), diffusione di luce (DLS) e scattering a basso e alto angolo dei raggi-X (SAXS/WAXS) risolti nel tempo, per lo studio della cinetica di formazione di un particolare sistema di interesse biotecnologico: i G-Hydrogel. I G-Hydrogel sono gel formati dalla combinazione per autoassemblaggio di molecole di Guanosina e GMP (Guanosina 5' monofosfato) in acqua che porta alla formazione di strutture chiamate quadruplessi, a loro volta capaci di formare una rete tridimensionale stabile che può assorbire grandi volumi d'acqua. Grazie alla loro biocompatibilità, questi gel sono utili in varie applicazioni, tra cui il trasporto di farmaci o l'immagazzinamento di proteine. In vista di un possibile uso di questo sistema nella local drug delivery che richiede la formazione del gel in situ a temperatura corporea, è stata studiata la cinetica di formazione e di propagazione del gel. I risultati ottenuti in laboratorio con POM e DLS mostrano come la formazione dei gel sia più veloce a basse concentrazioni d'acqua, mentre più lenta ad alte concentrazioni d'acqua. Allo stesso tempo tramite esperimenti di SAXS e WAXS svolti presso i sincrotroni Diamond Light Source di Oxford ed Elettra di Trieste è stato osservato che la struttura dei quadruplessi può variare in base al rapporto di Guanosina e GMP. Questi risultati sono importanti per comprendere come il gel si comporterebbe qualora si iniettassero separate soluzioni di Guanosina e GMP.
