This thesis examines the interaction between the gut microbiota and vitamin D, highlighting their role in maintaining intestinal and immune homeostasis. The gut microbiota performs essential metabolic, immune, and structural functions, whereas vitamin D, through the vitamin D receptor (VDR), regulates gene expression, contributes to the integrity of the intestinal epithelial barrier, and modulates the immune response. Current evidence suggests a bidirectional relationship between these two systems: vitamin D influences the composition of the gut microbiota, while the microbiota can modulate vitamin D metabolism. Alterations in VDR signaling have been associated with dysbiosis, increased intestinal permeability, and changes in the production of antimicrobial peptides. Furthermore, vitamin D promotes immune tolerance by enhancing the expansion of regulatory T cells and reducing pro-inflammatory immune responses, whereas the gut microbiota interacts with the mucosal immune system through the gut-associated lymphoid tissue (GALT). Experimental and observational studies support an association between vitamin D status and gut microbiota composition, although further research is needed to clarify the causal mechanisms underlying this relationship. Overall, the available evidence indicates that the vitamin D–VDR system and the gut microbiota constitute an integrated functional axis involved in maintaining intestinal and immune homeostasis.
Il presente elaborato analizza l’interazione tra il microbiota intestinale e la vitamina D, evidenziandone il ruolo nel mantenimento dell’omeostasi intestinale e immunitaria. Il microbiota intestinale svolge funzioni metaboliche, immunitarie e strutturali fondamentali, mentre la vitamina D, attraverso il recettore VDR, regola l’espressione genica, contribuisce all’integrità della barriera epiteliale intestinale e modula la risposta immunitaria. Le evidenze disponibili indicano l’esistenza di una relazione bidirezionale tra i due sistemi: la vitamina D influenza la composizione del microbiota intestinale, mentre il microbiota può modulare il metabolismo della vitamina D. Alterazioni del segnale VDR sono associate a disbiosi, aumento della permeabilità intestinale e modificazioni nella produzione di peptidi antimicrobici. Inoltre, la vitamina D favorisce la tolleranza immunitaria promuovendo l’espansione delle cellule T regolatorie e riducendo le risposte pro-infiammatorie, mentre il microbiota interagisce con il sistema immunitario mucosale attraverso il GALT. Studi sperimentali e osservazionali supportano l’esistenza di un’associazione tra stato vitaminico D e composizione del microbiota, sebbene siano necessari ulteriori studi per chiarirne i meccanismi causali. Nel complesso, il sistema vitamina D–VDR e il microbiota intestinale costituiscono un asse funzionale integrato coinvolto nel mantenimento dell’omeostasi dell’ospite.
