The aim of this thesis is to evaluate Adaptive Laboratory Evolution (ALE) as a strategy to address the challenges associated with the increase in grape sugar content resulting from rising temperatures. The higher sugar content of grapes leads to musts with a high potential alcohol content, and the ethanol produced during alcoholic fermentation exerts a toxic effect on the yeast cells that produce it, compromising the normal progression of the fermentation process and causing defects in the final product. For this reason, ALE represents a promising strategy, as it exploits the natural ability of yeasts to adapt to stressful conditions, enabling the selection of more resistant strains capable of completing fermentation efficiently. This work analyses the fermentative performance of evolved strains in comparison with their parental strains and investigates the molecular mechanisms underlying these improvements. Finally, the most recent technologies for the improvement of wine yeasts are briefly described, highlighting their potential to meet the new production and market demands of the wine industry.
L’obiettivo di questa tesi è valutare l’Adaptive Laboratory Evolution come strategia per affrontare la problematica dell’aumento del contenuto zuccherino dell’uva dovuto all’innalzamento delle temperature. L'elevato contenuto zuccherino delle uve determina mosti ad alto potenziale alcolico, l’etanolo prodotto a seguito della fermentazione risulta tossico per i lieviti che lo hanno generato compromettendo il regolare svolgimento della fermentazione e causando difetti al prodotto finale. Per questo motivo l’ALE risulta una strategia interessante in quanto sfrutta la capacità naturale dei lieviti di adattarsi a condizioni di stress per produrre ceppi più resistenti in grado di completare la fermentazione in maniera regolare. In questo lavoro analizzeremo la cinetica fermentativa dei ceppi evoluti rispetto a quelli parentali e quindi cercheremo di capire i meccanismi molecolari alla base di questi miglioramenti. Alla fine, descriveremo in breve le nuove tecnologie enologiche più recenti per andare incontro alle nuove esigenze produttive e di mercato.
