Among the biotic agents threatening wine production, Flavescence dorée represents a major threat to the entire sector, leading to the eradication of infected plants, for which no curative treatments are currently available. The only available strategies involve the control of the insect vector Scaphoideus titanus, which, through its feeding activity, is able to acquire and transmit this phytoplasma to other susceptible grapevines. Genetic transformation represents a biotechnology tool of particular interest for the introduction of agronomically relevant traits into grapevine, but its application is limited by the complexity of transformation protocols and by the strong influence of genotype on in vitro transformation and regeneration capacities. In particular, the use of genetically transformed rootstocks may be of interest within transgrafting strategies, making the development of efficient and reproducible transformation protocols for specific rootstock genotypes necessary. Therefore, the present work aimed to optimize the genetic transformation protocol of somatic embryos of the 110 Richter rootstock (Vitis berlandieri × V. rupestris) by modulating the duration of co-cultivation with Agrobacterium tumefaciens. For this purpose, somatic embryos were subjected to three different co-cultivation periods, namely 3, 4 and 5 days, using the EHA101 strain of A. tumefaciens containing the construct designed to silence the ATP2 gene of Scaphoideus titanus through RNA interference. Following co-cultivation with A. tumefaciens, the embryos were transferred to selection media based on MS salts at full or half strength and monitored over a period of 15 weeks, evaluating GFP expression, the occurrence of necrosis and bacterial contamination. Monitoring GFP expression allowed the influence of co-cultivation duration on the evolution of transformed tissues during the subsequent selection and regeneration period to be assessed. The results were also analyzed in relation to the occurrence of necrosis and the presence of A. tumefaciens contamination, highlighting a relationship between increased co-cultivation duration and greater exposure of the embryos to the effects associated with the presence of the bacterium. The comparison between full-strength MS and half-strength MS media also allowed the response of the embryos to different nutritional conditions during the selection and regeneration phases to be evaluated, establishing, under the experimental conditions adopted, that reducing the salt concentration resulted in a higher occurrence of necrosis in the explants in culture. The integration of the target sequence into the plant genome was finally verified by PCR on the regenerated lines subjected to molecular analysis. Overall, the work allowed the regeneration and obtaining of two genetically modified lines of the 110 Richter rootstock, defining a 4-day co-cultivation period as the most efficient in terms of the expression of GFP-positive explants and, consequently, of the number of independent lines obtained. Therefore, co-cultivation duration was confirmed as a key parameter in the optimization of the genetic transformation of the 110 Richter rootstock.

OPTIMIZATION OF AGROBACTERIUM TUMEFACIENS-MEDIATED GENETIC TRANSFORMATION OF 110 RICHTER GRAPEVINE ROOTSTOCK SOMATIC EMBRYOS BY MODULATING CO-CULTIVATION DURATION
