Long noncoding RNAs (lncRNAs) are transcripts longer than 200 nucleotides that do not encode proteins, but regulate numerous cellular processes through modulation of transcription, chromatin, and RNA and protein activity. In the oncology field, some lncRNAs contribute to the maintenance of cancer stem cells (CSCs), promoting their self-renewal, plasticity, invasiveness and resistance to treatments. Their altered expression can therefore support tumor progression and the onset of recurrences. This report explored three strategies for the therapeutic targeting of lncRNAs. Antisense oligonucleotides (ASOs) bind to the target transcript and promote its degradation; CRISPRi uses an inactivated Cas9 to reduce its transcription without cutting DNA; Cas13d instead acts directly on the RNA already produced. The three technologies differ in mechanism of action, duration of effect, specificity, and mode of administration. Although preclinical studies show promising results, clinical application requires more accurate target selection, reduction of adverse effects, and delivery systems capable of selectively reaching the tumor and CSCs.

Abstract
Although preclinical studies show promising results, clinical application requires more accurate target selection, reduction of adverse effects, and delivery systems capable of selectively reaching the tumor and CSCs.| File | Dimensione | Formato | |
