With this thesis i want to demonstrate the importance of post-transcriptional mechanisms, emphasizing the importance of RBP, in particular the role of HuR in neurodevelopment and especially in noeocortogenesis. Demonstrating that it specifically binds mrna of Foxp1 and Foxp2 transcription factors associated with autism spectrum pathologies. This link takes place according to a precise temporality, it has been evidenced that such regulation derives from an inhibition of the translation obtained through the binding with the 3'-UTR regions of the transcripts. It has also been highlighted as the differential phosphorylation of some sites on HuR leads to the regulation of its activity. In addition to this it has been seen that HuR is only one of the many RBPs involved in neurodevelopment, which form a very heterogeneous group of proteins with different targets and which all lead to a temporal and spatial regulation of protein synthesis. In addition to this, for a more complete view of the complex mechanism behind the development of the neocortex, i briefly analyzed other molecules involved in this development namely : complexes of repression of synthesis, IncRNA, miRNA. Finally, to underline the importance that all the processes described lead to a fine post-transcriptional regulation, some pathologies connected with incorrect post-transcriptional mechanisms have been analyzed.

Con questa tesi si è voluto dimostrare l'importanza dei meccanismi post-trascrizionali, sottolineando l'importanza delle RBP, in particolare il ruolo di HuR nel neurosviluppo e soprattutto nella noeocortogenesi. Dimostrando che lega specificamente gli mRNA dei fattori di trascrizione Foxp1 e Foxp2 associati a patologie dello spettro autistico. Questo legame avviene a seconda di una precisa temporalità, si è evidenziato che tale regolazione deriva da una inibizione della traduzione ottenuta tramite il legame con le regioni 3'-UTR dei suddetti trascritti. Inoltre è stato messo in luce come la fosforilazione differenziale di alcuni siti su HuR porti alla regolazione della sua attività. In aggiunta a ciò è stato visto che HuR é solo una delle molteplici RBPs coinvolte nel neurosviluppo, che formano un gruppo molto eterogeneo di proteine con diversi target e che portano tutte ad una regolazione temporale e spaziale della sintesi proteica. In aggiunta a ciò, per una visione più completa del complesso meccanismo alla base dello sviluppo della neocorteccia, sono state brevemente analizzate altre molecole implicate in questo sviluppo ovvero : complessi di repressione della sintesi, IncRNA, miRNA. Infine per sottolineare l'importanza che tutti i processi descritti conducano ad una fine regolazione post-trascrizionale sono state analizzate alcune patologie connesse con errati meccanismi post-trascrizionali.

Il ruolo di HuR e di altre "RNA Binding Proteins" nei meccanismi post-trascrizionali coinvolti nel neurosviluppo in condizioni fisiologiche e patologiche.
